Important Safety Information regarding Lenzetto 1.53 mg/spray Transdermal Spray

First of all, thank you for choosing Numan. If your prescription request for Lenzetto has been accepted, or if you considering whether this treatment may be suitable for you, we'd like to help you learn how to use it to its best effect, and give you some important information before you start taking it.

What is it?

Lenzetto 1.53 mg/spray Transdermal Spray is an oestrogen-only hormone replacement therapy (HRT) that is delivered as a transdermal (through the skin) spray, and in this way is also absorbed directly into the bloodstream. Its active ingredient is estradiol (as estradiol hemihydrate), which is a body-identical form of the female hormone oestrogen, meaning it is chemically the same as the oestrogen the body produces naturally.

What is it used for?

Lenzetto is used for the relief of postmenopausal symptoms such as hot flushes, night sweats, mood swings, and vaginal dryness.

Progestogen Requirement: If you have a uterus, you must also take a separate progestogen to protect your womb lining. Speak to your healthcare provider for further advice if you are unsure.

How long does it take to work?

Many women notice an improvement in their symptoms within a few weeks of starting treatment. However, it can sometimes take up to three months for your body to adjust and for you to feel the full benefits.

How do I use it?

Before using a new applicator for the first time, you must prime it by spraying three times into the cap. This only needs to be done once per device. The spray should be applied once daily, at the same time each day.

To apply a dose:

  • Hold the applicator upright against a clean, dry, and unbroken area of skin on your inner forearm

  • Press the button once fully to apply a dose.

  • If your prescribed dose is more than one spray, apply the subsequent sprays to adjacent areas of the forearm, or on the inner thigh, making sure the sprays do not overlap.

  • Allow the spray to dry for at least 2 minutes before getting dressed, and do not wash, swim, or bathe for at least 60 minutes after application.

You should also use Lenzetto with caution in extreme temperature conditions, such as sun bathing or saunas. Speak to your healthcare provider for further advice if you are unsure.

Missed dose: If a dose is missed, you should make up for the missed dose as soon as you remember, and take the next dose at the usual time. If it is almost time for the next dose, you should skip the missed dose and take the next dose at the usual time. If one or more doses are missed, one primer spraying with the cover on is needed. Forgetting a dose may increase the likelihood of breakthrough bleeding and spotting.

If you do miss one or more doses, prepare the applicator according to the

instructions given in your Patient Information Leaflet (linked at the bottom of this page) under the section “If you forget to use Lenzetto”. Always take this medicine exactly as instructed, and check with your healthcare provider if you are not sure.

Special dosing requirements

The usual starting dose of Lenzetto is one spray per day. If your symptoms do not improve after at least four weeks, your clinician may increase your dose to two, or a maximum of three, sprays per day. The goal of HRT is always to use the lowest effective dose for the shortest duration necessary to manage your symptoms.

There are no special dosage requirements for elderly patients, though experience in treating women over 65 years is limited. This medicine is not for use in children.

Overweight and obese women: There is some limited data that the rate and extent of absorption of Lenzetto can be reduced in overweight and obese women. During your treatment, the dose of Lenzetto may require adjustment by your clinician if needed.

Information on fertility, pregnancy and lactation

Pregnancy: Lenzetto is not a contraceptive and is intended for use only in post-menopausal women. If it is less than 12 months since your last menstrual period or you are under 50 years old, you may still need to use additional contraception to prevent pregnancy. Speak to your healthcare provider for advice on this. If you become pregnant while taking it, you must stop the medication immediately and contact your healthcare provider.

Breastfeeding: Lenzetto is not indicated during breastfeeding.

Other important safety information

It is crucial to understand that if you have not had a hysterectomy (i.e., you still have your womb), you must take a separate progestogen hormone alongside Lenzetto. This is typically prescribed for 12 to 14 days each month. This is a vital safety measure to protect the lining of your womb from the effects of oestrogen, which, when taken alone, can lead to a build-up of the lining (endometrial hyperplasia) and increase the risk of endometrial cancer.

Risk of Transference: The hormone in the Lenzetto spray can be accidentally transferred to other people through skin-to-skin contact. It is very important to prevent others, especially children and pets, from coming into contact with the application area. You should cover the treated area with clothing once the spray is dry and avoid contact for at least 60 minutes. There have been reports of children showing unexpected signs of puberty (like breast budding) after accidental exposure. If you suspect someone has been exposed, wash their skin with soap and water immediately and consult a doctor.

Flammability: Lenzetto is alcohol-based and is flammable. You must avoid fire, open flames, or smoking while applying the spray and until it has completely dried on your skin.

What are the associated risks with treatment?

Common side effects of treatment with Lenzetto include skin irritation where the spray is applied, headache, abdominal pain, nausea, breast tenderness, and irregular vaginal bleeding. As a transdermal form of HRT, Lenzetto bypasses the liver and carries a lower risk of blood clots (venous thromboembolism) compared to other systemic forms of HRT, such as oral tablets or capsules. The other risks associated with systemic HRT, such as a small increased risk of breast cancer and stroke, still apply, although the overall risk remains low for most patients being treated. What monitoring or follow-up is needed to continue taking treatment safely?

Your clinician will review your treatment after three months to check how you’re responding. After that, you should have a check-up at least once a year. These reviews will assess whether HRT is still right for you, make sure your symptoms are under control, ask about any side effects and vaginal bleeding, request a check of your weight and your blood pressure, and decide if you need further tests such as a bone density scan. You should also examine your breasts regularly, and make sure to attend all routine screening appointments such as mammograms and cervical screening, if applicable to you. If you have a premature menopause, the risks of using HRT may be different, which you should discuss with your clinician.

When to stop treatment

You should stop using Lenzetto immediately and consult your doctor if you experience any of the following: signs of a blood clot (e.g. painful swelling in the leg), a swollen face, tongue and/or throat and/or difficulty swallowing or hives, together with difficulty breathing which are suggestive of an angioedema, jaundice (yellowing of the skin or eyes), a significant increase in your blood pressure, the new onset of a migraine-type headache, or if you become pregnant. When you and your clinician decide it is time to stop HRT, the dose is often reduced gradually to help prevent your menopausal symptoms from returning suddenly.

Conditions which need supervision

If you have any of the following conditions, or if you’ve had them before, especially if they worsened during pregnancy or previous hormone treatment, you should be closely monitored while taking Lenzetto 1.53 mg/spray Transdermal Spray. These conditions may return or become worse during treatment, particularly:

  • Leiomyoma (uterine fibroids) or endometriosis

  • Risk factors for thromboembolic disorders

  • Risk factors for oestrogen dependent tumours, e.g. 1st degree heredity for breast cancer

  • Hypertension

  • Liver disorders (e.g. liver adenoma)

  • Diabetes mellitus with or without vascular involvement

  • Cholelithiasis

  • Migraine or (severe) headache

  • Systemic lupus erythematosus

  • A history of endometrial hyperplasia

  • Epilepsy

  • Asthma

  • Otosclerosis

Storage instructions

Do not refrigerate or freeze. Do not store above 25°C. Use within 56 days of first use. Store away from heaters, open flames, and other sources of ignition. Keep out of reach of children.
